Many system files and "in-use" files cannot be moved/defragmented.

It is best to ONLY defragment after performing a full system cleanup (virus
checks, application uninstalls, spyware scans and a CHKDSK) and rebooting.
Then - after turning off your screensaver and anything else that may access
the drive (close all windows and applications - exit ones in the taskbar -
etc) - defragment.

Is all that "necessary"? No.
Will it give you more consistent and reliable results? Probably.
